采用PCR技术克隆得到12尾巨魾(Bagarius yarrelli)的细胞色素氧化酶Ⅰ(COⅠ)、细胞色素氧化酶Ⅱ(COⅡ)和细胞色素氧化酶Ⅲ(COⅢ)的基因序列。结果显示:巨魾COⅠ基因序列全长1 551 bp,12个个体共出现9个单倍型,13个突变位点;COⅡ基因序列全长691 bp,12个个体共出现5个单倍型,5个突变位点;COⅢ基因序列全长784 bp,12个个体出现7个单倍型,8个突变位点。通过最小进化法Minimum Evolution(ME)构建系统发育树分析显示,巨魾与14种鮡科鱼在序列上有一定的同源性,并且COⅠ、COⅡ和COⅢ都与中华纹胸鮡(Glyptothorax sinense)、福建纹胸鮡(Glyptothorax fukiensis)、三线纹胸鮡(Glyptothorax trilineatus)和长丝黑鮡(Gagata dolichonema)在同一个分支,表明巨魾与纹胸鮡属和黑鮡属有较近的亲缘关系。
The gene sequences of cytochrome oxidase Ⅰ (CO Ⅰ), cytochrome oxidase Ⅱ (CO Ⅱ) and cytochrome oxidase Ⅲ (CO Ⅲ) were isolated from 12 giant-sized Bagarius yarrelli by PCR. The results showed that the sequence of COⅠ gene was 1 551 bp in length, 9 haplotypes and 13 mutation sites were found in 12 individuals, the COⅡ gene sequence was 691 bp in length, 5 haplotypes were found in 12 individuals, 5 mutated sites; CO Ⅲ gene sequence length of 784 bp, 12 individuals appear 7 haplotypes, 8 mutation sites. The Phylogenetic tree analysis of Minimum Evolution (ME) by Minimum Evolution (ME) showed that there were some homologies in the sequence between giant scodoptera and scythetic fish, and COⅠ, COⅡ and COⅢ were closely related to Glyptothorax sinense, Glyptothorax fukiensis, Glyptothorax trilineatus and Gagata dolichonema are all on the same branch, indicating that Jujubes are closely related to Pleurotus ostreatus and Hemiptera .
