目的研究白血病患者的骨髓血管新生程度和骨髓液血管内皮生长因子(VEGF)水平的相关性。方法采用EnVison免疫组化法,用兔抗人vWF(vonWille蛳brandfactor)多克隆抗体标记骨髓内皮细胞;采用ELISA法检测骨髓液中VEGF的含量。结果骨髓MVD在各实验组均明显高于正常对照组(P<0.05)。骨髓液VEGF水平在AML、CML组明显高于正常对照组(P<0.01),ALL组与正常对照组比较差异无显著性(P>0.05)。对各白血病组的MVD和VEGF含量进行了相关性分析,结果表明在AML或CML组中VEGF水平和MVD呈正相关性(分别为r=0.648,P<0.05;r=0.733,P<0.05)。结论白血病患者骨髓微血管数增多,提示血管新生在白血病发病中有重要作用,VEGF可能在一定程度上对血管新生有促进作用。
Objective To investigate the correlation between bone marrow angiogenesis and bone marrow vascular endothelial growth factor (VEGF) levels in patients with leukemia. Methods EnVison immunohistochemistry was used to label bone marrow endothelial cells with polyclonal antibody against vWF (von Willebrand factor). The content of VEGF in bone marrow fluid was detected by ELISA. Results The bone marrow MVD in each experimental group was significantly higher than that in the normal control group (P <0.05). Bone marrow VEGF levels in AML, CML group was significantly higher than the normal control group (P <0.01), ALL group and the normal control group showed no significant difference (P> 0.05). The correlation analysis of MVD and VEGF in each leukemia group showed that there was a positive correlation between VEGF and MVD in AML or CML group (r = 0.648, P <0.05; r = 0.733, P <0.05). Conclusion The increase of bone marrow microvessels in leukemia patients suggests that angiogenesis plays an important role in the pathogenesis of leukemia. VEGF may promote angiogenesis to a certain extent.
