目的:探讨不同温度热疗后宫颈癌CaSki细胞中Survivin的表达。方法:采用实时荧光定量PCR技术和蛋白质印迹法,检测45℃、47℃实验组和37℃对照组宫颈癌CaS-ki细胞中Survivin的表达。结果:SurvivinmRNA在37℃、45℃和47℃处理后24 h的表达倍数分别为1、0.707±0.089和0.814±0.068,F=114.48,P<0.001。Survivin蛋白在对照组呈高表达,在45℃和47℃的表达明显减弱甚至完全缺失,F=14 355.4,P<0.001。结论:>45℃的热疗可以在mRNA的水平抑制宫颈癌CaSki细胞中Survivin表达,导致Survivin蛋白表达降低甚至完全缺失。
Objective: To investigate the expression of Survivin in cervical cancer CaSki cells after different temperature hyperthermia. Methods: Real-time fluorescent quantitative PCR and Western blotting were used to detect the expression of Survivin in cervical cancer CaS-ki cells at 45 ℃, 47 ℃ and 37 ℃. Results: The expression levels of Survivin mRNA at 24 h after treatment with 37 ℃, 45 ℃ and 47 ℃ were 1.707 ± 0.089 and 0.814 ± 0.068, respectively, F = 114.48, P <0.001. Survivin protein was highly expressed in the control group, and its expression was significantly weakened or even completely disappeared at 45 ℃ and 47 ℃, F = 14 355.4, P <0.001. CONCLUSION: Hypothermia> 45 ℃ can inhibit the expression of Survivin in cervical cancer CaSki cells at the mRNA level, resulting in the reduction or even absence of Survivin protein expression.
