目的探讨原位逆转录 -聚合酶链反应技术 (ISRT -PCR)在普通制作石蜡包埋组织中的应用及价值。方法应用直接法ISRT -PCR方法对普通制作、石蜡包埋淋巴瘤组织中的丙型肝炎病毒 (HCV)基因组序列进行检测。结果从存档多年的福尔马林固定、普通制作石蜡包埋淋巴瘤组织中检测到HCVRNA序列。结论ISRT -PCR可用于福尔马林固定、普通制作石蜡包埋组织中低拷贝目的基因组序列的检测方法为常规制作的存档石蜡包埋组织的充分利用提供了一条新的途径
Objective To investigate the value and application of in situ reverse transcription-polymerase chain reaction (ISRT-PCR) in normal paraffin-embedded tissues. Methods Direct sequencing method ISRT-PCR was used to detect the genome sequence of hepatitis C virus (HCV) in paraffin-embedded lymphoma tissues. Results HCVRNA sequences were detected from formalin-fixed, normal-made paraffin-embedded lymphoma tissue, which was archived for many years. Conclusion ISRT-PCR can be used in formalin fixation. The detection method of low-copy genomic sequence in common paraffin-embedded tissue provides a new way for the routine use of archived paraffin-embedded tissue
