目的:了解动脉平滑肌细胞(ASMC)的蛋白激酶C(PKC)含量及亚型分布,以及与细胞增殖的关系。方法:采用点印迹技术及MTT方法,检测了人ASMC颗粒部分与胞浆部分PKC的6种亚型的含量和分布。结果:①人ASMC细胞中主要有PKCα、PKCβ、PKCζ3种亚型分布,并发现63%分布在胞浆中;②人ASMC细胞PKC总量随着佛波酯(PMA)、胰岛素浓度的增加而增加,人ASMC细胞增加数量也随着PMA、胰岛素的浓度增加而增加。结论:人ASMC细胞中以PKCα、PKCβ、PKCζ3型含量较多,胰岛素可能是通过激活PKC信号传导途径引起细胞的基因表达增高,导致ASMC细胞增殖。
Objective: To investigate the content and subtype distribution of protein kinase C (PKC) in arterial smooth muscle cells (ASMCs) and its relationship with cell proliferation. Methods: The content and distribution of 6 subtypes of PKC in the granular and cytoplasmic fractions of human ASMC were detected by dot blot technique and MTT assay. Results: (1) There were three subtypes of PKCα, PKCβ and PKCζ in human ASMC cells, and 63% were found in the cytoplasm. (2) The total amount of PKC in human ASMC cells increased with the increase of PMA and insulin concentration Increasing, the number of human ASMC cells increased with the PMA, insulin concentration increased. CONCLUSION: The content of PKCα, PKCβ and PKCζ3 in human ASMC cells is higher than that in control group. Insulin may increase the gene expression of cells through activation of PKC signal transduction pathway, leading to the proliferation of ASMC cells.
