目的:观察血栓素(TX)A2合成酶抑制剂OKY-046对嗜酸性粒细胞(EOS)浸润气道的影响,探讨TXA2在过敏性气道炎症过程中的作用。方法:应用鸡卵白蛋白(OVA)致敏和反复雾化吸入刺激小鼠以复制过敏性气道炎症模型。除对照组外,各实验组小鼠于每天OVA刺激前分别给予不同剂量OKY-046腹腔注射,计数支气管肺泡灌洗液(BALF)中EOS数量,并测定支气管肺泡灌洗液(BALF)中TXB2和6-酮-前列腺素F1α(6-Keto-PGF1α)水平。结果:致敏小鼠给予OVA刺激后BALF中EOS数、TXB2和6-Keto-PGF1α水平均显著升高,当以OKY-046处理后,BALF中的EOS明显减小,并呈剂量相关性,而且OKY-046尚能引起TXB2水平呈剂量相关性降低。结论:TXA2对于EOS浸润到呼吸道具有明显的趋化作用,其合成酶抑制剂对于临床治疗哮喘患者可能会有重要的价值
Objective: To observe the effect of TXY2 synthase inhibitor OKY-046 on eosinophil infiltration in airway and to explore the role of TXA2 in allergic airway inflammation. Methods: Mice were stimulated with ovalbumin (OVA) sensitization and repeated inhalation to replicate the allergic airway inflammation model. Except for the control group, the mice in each experimental group were given different doses of OKY-046 intraperitoneally before OVA stimulation every day, and the number of EOS in bronchoalveolar lavage fluid (BALF) was counted. The levels of TXB2 in bronchoalveolar lavage fluid (BALF) And 6-keto-prostaglandin F1α (6-Keto-PGF1α) levels. Results: The levels of EOS, TXB2 and 6-Keto-PGF1α in BALF of sensitized mice were significantly increased after OVA stimulation. When OKY-046 was treated, the EOS of BALF was significantly reduced and the dose-dependent, And OKY-046 still can cause dose-related reduction TXB2 level. Conclusion: TXA2 has obvious chemotactic effect on the infiltration of EOS into the respiratory tract, and its synthetase inhibitor may have important value in the clinical treatment of patients with asthma
