目的 六味地黄多糖 C A43 B 和 P3 是具有免疫调节作用的活性多糖,在此基础上进一步观察其体外对肿瘤细胞增殖的抑制作用。方法 用[3 H] Td R 参入法,琼脂克隆形成法及细胞形态学和 N B T 还原染色等方法观察其对小鼠淋巴细胞白血病细胞系 L1210 、人早幼粒白血病细胞系 H L60增殖反应、克隆存活率的影响以及对 H L60 细胞的分化诱导作用。结果 C A43 B和 P3 对 L1210 和 H L60 细胞的体外增殖反应和克隆形成率具有不同程度的抑制作用,对 H L60细胞的分化形态及 N B T 阳性细胞率无明显影响,但可以使 H L60 细胞体积明显缩小。结论 六味地黄多糖 C A43 B 和 P3 对肿瘤细胞生长有抑制作用,可能是具有免疫调节和直接抑瘤双重作用的活性多糖;另外提示 C A43 B 和 P3 对 H L60 细胞分化有一定的诱导作用
Objective Polysaccharides of Rehmannia glutinosa C A4 3 B and P 3 are active polysaccharides with immunomodulatory effects. On this basis, their inhibitory effects on tumor cell proliferation were further observed. Methods The proliferation of mouse lymphocytic leukemia cell line L1210 and human promyelocytic leukemia cell line H L60 was observed by [3 H] Td R incorporation method, agar cloning method, cell morphology and N B T reductive staining. Response, clonal survival, and differentiation induction of HL60 cells. RESULTS: C A4 3 B and P 3 had different degrees of inhibitory effect on the in vitro proliferative response and colony forming rate of L1210 and H L60 cells, but had no significant effect on the differentiation morphology and N B T positive cell rate of H L60 cells. Can significantly reduce the volume of H L60 cells. Conclusion The polysaccharides of A. rehmannia stalks C A4 3 B and P 3 can inhibit the growth of tumor cells, and may be active polysaccharides with dual functions of immune regulation and direct inhibition of tumors.
