目的建立癫痫患者脑组织差异表达的蛋白质谱,进一步阐明癫痫的发病机制、寻找新的治疗靶点。方法采用双向荧光差异凝胶电泳(2D-DIGE)和基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)对癫痫患者致痫灶脑组织差异表达的蛋白质进行分析鉴定。结果在患者致痫灶脑组织中共鉴定出15个蛋白质点出现1.5倍以上表达差异,其中13个下调,2个上调。结论癫痫患者脑组织中存在多个差异表达的蛋白质,可能与癫痫的发病机制相关。
Objective To establish a protein spectrum differentially expressed in brain tissue of patients with epilepsy, further clarify the pathogenesis of epilepsy and find new therapeutic targets. Methods Proteins differentially expressed in epileptic foci of epileptic patients were identified by 2D-DIGE and MALDI-TOF-MS. Results A total of 15 protein spots were identified more than 1.5 times in the epileptic foci of patients, of which 13 were down-regulated and 2 were up-regulated. Conclusion There are many differentially expressed proteins in the brain tissue of epileptic patients, which may be related to the pathogenesis of epilepsy.
