以体外细胞培养和 MTT 比色法,比较经不同浓度甲酰四氢叶酸钙(CF)调变后5-氟脲嘧啶(FU)对大肠癌细胞株 SW-1116的抑制效应。结果表明,当 FU 浓度达到200μg/ml 并作用72小时后,方见 CF 对 FU 有明显的增效作用;而此种增效作用在 CF 浓度为120μg/ml 和12μg/ml 之间无显著差异。以上提示,(1)为达致增效之目的,FU 的临床剂量和静脉滴注时间应以能维持较高、较久的血浓度为宜;(2)CF 对 FU 增效的剂量阈值尚待确定,为减轻毒副反应,目前所取的大剂量 CF 可望酌减,而快速滴注的效果则尚待证实。
The inhibitory effect of 5-fluorouracil (FU) on colorectal cancer cell line SW-1116 after different concentrations of calcium folinate (CF) was regulated by in vitro cell culture and MTT colorimetric assay. The results showed that when FU concentration reached 200 μg/ml and the effect was 72 hours, Fang-Feng had a significant synergistic effect on FU; however, there was no significant difference in this synergism between CF concentrations of 120 μg/ml and 12 μg/ml. . The above suggested that: (1) To achieve the purpose of increasing efficiency, the clinical dosage and intravenous infusion time of FU should be maintained at a higher and longer blood concentration; (2) the dose threshold for the efficacy of CF on FU is still satisfactory. To be determined, in order to reduce toxic side effects, the large doses of CF currently taken are expected to be reduced, while the effect of rapid instillation remains to be confirmed.
