目的:探讨藏药巴桑母酥油丸对放射线--化学复合损伤小鼠红系造血功能的影响及可能机制。方法:采用外周血红细胞计数、造血祖细胞集落分析、实时荧光定量多聚酶链反应等方法,检测灌胃巴桑母酥油丸(0.75,1.5,3.0 g·kg-1)后放射线-化学复合损伤小鼠外周血红细胞(RBC)数及血红蛋白(Hb)量、骨髓红系造血祖细胞集落产率、肾脏红细胞生成素(Epo)和脾脏红细胞生成素受体(EpoR)mRNA表达量。结果:灌胃14,20 d后,巴桑母酥油丸中、高剂量组外周血RBC数显著高于空白组、模型组和低剂量组(P<0.05);灌胃20 d后,巴桑母酥油丸中剂量Hb含量显著高于空白组和模型组(P<0.05)。灌胃14 d后,巴桑母酥油丸中剂量组骨髓细胞的早期、晚期红系祖细胞(CFU-E,BFU-E)集落产率均显著高于空白组和模型组(P<0.01);中剂量组肾脏Epo mRNA相对表达量显著高于空白组和模型组(P<0.05)。结论:巴桑母酥油丸可以通过刺激肾脏Epo分泌、促进骨髓造血祖细胞增殖分化,进而促进放射线-化学复合损伤小鼠外周血RBC数及Hb含量的恢复。
Objective: To investigate the effect and possible mechanism of Tibetan Pancen Chaxue pill on erythrocytic hematopoiesis in radiation-chemical injury mice. Methods: Peripheral blood erythrocyte count, hematopoietic progenitor cell colony analysis and real-time fluorescence quantitative polymerase chain reaction were used to detect the radiation-chemical complex damage after oral administration of Pancenzhan Gum Oil Pills (0.75,1.5 and 3.0 g · kg-1) (RBC), hemoglobin (Hb), hematopoietic progenitor cell yield, erythropoietin (Epo) and spleen erythropoietin receptor (EpoR) mRNA expression in bone marrow. Results: After 14 and 20 days of intragastric administration, the peripheral blood RBC counts of middle and high dose group of Pancen mother shortening pills were significantly higher than those of blank group, model group and low dose group (P <0.05) The content of Hb in the middle shortening pills was significantly higher than that in the blank group and the model group (P <0.05). After 14 days of intragastric administration, the yield of early and late erythroid progenitor cells (CFU-E, BFU-E) in the medium-dose group of Psoriasis Glabra pills was significantly higher than that of the blank group and the model group (P <0.01) ; The relative expression of Epo mRNA in middle dose group was significantly higher than that in blank group and model group (P <0.05). Conclusion: Psoriasis Glabraum Pill can promote the proliferation and differentiation of hematopoietic progenitor cells of bone marrow by stimulating the secretion of Epo in kidney, and then promote the recovery of RBC number and Hb content in peripheral blood of mice with radiation - chemical compound injury.
