目的研究丹参饮片高效液相(HPLC)指纹图谱,建立不同级别丹参饮片HPLC指纹图谱及其分级方法。方法运用戴安U3000-DAD-HPLC色谱仪,采用依利特C-18BP(4.6mm×250mm,5μm)柱,以0.8%冰乙酸的甲醇溶液-0.8%冰乙酸水溶液梯度洗脱,检测波长270nm,柱温30℃。采用聚类分析和指纹图谱相似度评价方法分析24批丹参饮片的指纹图谱。结果 24批丹参饮片获得15个共有峰,相似度在0.8以上,样品聚类将饮片分为3类,聚类结果与峰面积比值相关联。结论该方法可作为丹参饮片内在质量评价的依据之一。
Objective To study HPLC fingerprints of Salvia miltiorrhiza radix salviae miltiorrhizae Pieces and to establish HPLC fingerprints of Salvia miltiorrhiza radix Pieces of different grades and their grading methods. Methods A Dionex U3000-DAD-HPLC chromatograph was used to elute gradient elution with a gradient of eluent C-18BP (4.6 mm × 250 mm, 5 μm) with 0.8% glacial acetic acid in methanol-0.8% Column temperature 30 ℃. The fingerprints of 24 batches of Salvia miltiorrhiza were analyzed by clustering analysis and fingerprint similarity evaluation. Results 24 batches of Salvia miltiorrhiza tablets obtained 15 common peaks, the similarity was above 0.8. The samples were divided into three groups according to the sample clustering. The clustering results were correlated with the peak area ratio. Conclusion This method can be used as one of the bases for the internal quality evaluation of Danshen Pieces.
