目的 通过原代培养的正常及受H2 O2 损伤的肺泡Ⅱ型上皮细胞 (ATⅡcell) ,探讨异氟醚 (Iso)对肺泡Ⅱ型上皮细胞合成表面活性物质的影响。方法 肺泡Ⅱ型上皮细胞分离、纯化、培养 32h后 ,在正常细胞或 75 μmol·L-1H2 O2 处理细胞的培养液内加入Iso 0 2 8、2 8mmol·L-1,继续培养 2h。分别通过MTT法检测肺泡Ⅱ型上皮细胞增殖功能 ,3H 胆碱参入法测其合成表面活性物质的功能。结果 Iso降低正常肺泡Ⅱ型上皮细胞合成表面活性物质的功能并加重H2 O2 引起的肺泡Ⅱ型上皮细胞表面活性物质合成功能的降低 ,而Iso对正常及受 (H2 O2 )损伤的ATⅡcell增殖功能均无显著影响。结论 Iso可以降低培养的肺泡Ⅱ型上皮细胞合成表面活性物质的功能 ,尤其是在过氧化环境中。
Objective To investigate the effects of isoflurane (Iso) on the synthesis of surfactant in alveolar type Ⅱ epithelial cells by primary culture of normal and H2O2 injured alveolar type Ⅱ epithelial cells (ATⅡcell). Methods Alveolar epithelial type Ⅱ epithelial cells were isolated, purified and cultured for 32 h. Iso 0 2 8,2 8 mmol·L-1 was added into the culture medium of normal cells or 75 μmol·L-1 H2 O2-treated cells for 2 h. The proliferation of alveolar type Ⅱ epithelial cells was detected by MTT method and the function of synthetic surfactant was measured by 3H choline incorporation method. Results Iso reduced the function of surfactant synthesis in normal alveolar type Ⅱ epithelial cells and increased the H2O2-induced decrease in the synthesis of surfactant type II alveolar epithelial cells. However, Iso inhibited the proliferation of ATⅡcell induced by H2O2 No significant effect. Conclusion Iso can reduce the function of cultured alveolar type Ⅱ epithelial cells in the synthesis of surface-active substances, especially in the environment of peroxidation.
