目的:探讨前列腺癌(PCa)组织中缺氧诱导因子-1α(HIF-1α)和基质金属蛋白酶9(MMP-9)的表达及其临床意义。方法:应用免疫组织化学法检测了20例良性前列腺肥大(BPH),10例前列腺上皮内肿瘤(PIN)和56例PCa组织中HIF-1α和MMP-9的表达情况。结果:Pca组织中HIF-1α和MMP-9的表达均明显高于BHP和PIN(P<0.01),而后两者中HIF-1α、MMP-9的表达比较均无显著性差异(P>0.05)。Pca组织中HIF-1α和MMP-9的表达均与Gleason分级、临床分期、癌穿透被膜、骨转移及淋巴结转移有关(P<0.05),而与年龄及病变部位无关(P>0.05)。Pca组织中HIF-1α和MMP-9蛋白阳性表达呈正相关(rs=0.729,P<0.05)。结论:HIF-1α、MMP-9蛋白与前例腺癌的生长、侵袭、转移密切相关,HIF-1α可能通过上调MMP-9的表达来提高肿瘤细胞的侵袭力。
Objective: To investigate the expression of hypoxia inducible factor-1α (HIF-1α) and matrix metalloproteinase 9 (MMP-9) in prostate cancer (PCa) tissues and their clinical significance. Methods: The expressions of HIF-1α and MMP-9 in 20 cases of benign prostatic hypertrophy (BPH), 10 cases of prostatic intraepithelial neoplasia (PIN) and 56 cases of PCa were detected by immunohistochemistry. Results: The expressions of HIF-1α and MMP-9 in Pca tissues were significantly higher than those in BHP and PIN (P <0.01), while there was no significant difference in the expression of HIF-1α and MMP-9 ). The expression of HIF-1α and MMP-9 in Pca tissues was correlated with Gleason grade, clinical stage, cancer penetrating capsule, bone metastasis and lymph node metastasis (P <0.05), but not with age and location of lesions (P> 0.05). The positive expression of HIF-1α and MMP-9 in Pca tissues was positively correlated (rs = 0.729, P <0.05). Conclusion: HIF-1α and MMP-9 are closely related to the growth, invasion and metastasis of adenocarcinoma. HIF-1α may increase the invasiveness of tumor cells by up-regulating the expression of MMP-9.
