目的观察方格星虫粗提物(SNP)对5/6肾切除大鼠肾小球病变的影响。方法 SD大鼠40只,随机分为假手术组和手术组,手术组行5/6肾切除术并于术后2周随机分为模型组、SNP低剂量组、SNP高剂量组。在给药前及给药4、8、12、16 w末观察大鼠24 h尿蛋白(24 hUPQ)及血肌酐(Scr)的动态变化,普通光镜观察肾小球病理变化,免疫组化法检测肾组织中纤维连接蛋白(FN)、α-平滑肌肌动蛋白(-αSMA)、Wilm肿瘤蛋白-1(WT-1)的表达。结果随着时间的延长,各组大鼠24 hUPQ、Scr均逐渐升高,但同一时间点,治疗组与模型组比较,差异无统计学意义;治疗组大鼠肾小球系膜增生程度、FN及-αSMA表达均较模型组下降;治疗组WT-1表达与模型组比较差异不显著。结论方格星虫粗提物可减轻5/6肾切除大鼠肾小球系膜增生程度、其机制可能与下调FN、-αSMA的表达有关。
Objective To observe the effect of crude extract of Pleurotus citriclus on glomerular lesions in 5/6 nephrectomized rats. Methods Forty SD rats were randomly divided into sham operation group and operation group. The operation group was treated with 5/6 nephrectomy and divided randomly into model group, SNP low dose group and SNP high dose group at 2 weeks after operation. The changes of 24 h urinary protein (24 hUPQ) and serum creatinine (Scr) were observed before administration and at the end of 4, 8, 12 and 16 weeks. The pathological changes of glomeruli were observed with light microscope. Immunohistochemistry Method to detect the expression of FN, -α SMA and Wilm tumor protein-1 (WT-1) in renal tissue. Results With the extension of time, the rats in each group 24 hUPQ, Scr were gradually increased, but at the same time point, the treatment group and the model group, the difference was not statistically significant; the treatment group rats mesangial proliferation, FN and -αSMA expression were lower than the model group; WT-1 expression in the treatment group compared with the model group was no significant difference. Conclusion The crude extracts of Pleurotus citri can reduce the mesangial proliferation in 5/6 nephrectomized rats. The mechanism may be related to the down-regulation of FN and -αSMA expression.
