目的对海藻酸钙和卡波姆凝胶经支气管镜介入兔肺支气管内的安全性进行评价,为含抗结核药物凝胶肺内应用提供依据。方法 45只日本长耳兔随机分成对照组、海藻酸钙组和卡波姆组,每组15只,实验观测4周。检测体温、体重、血常规、T细胞亚群及肺病理学改变。结果各组间体温、体重、血常规及外周血T细胞亚群的比较均无显著性差异(P>0.05)。海藻酸钙组和卡波姆组凝胶介入前与介入后CD3+、CD4+、CD8+百分比比较亦无显著性差异(P>0.05)。海藻酸钙组和卡波姆组的受试动物左肺底细支气管无凝胶成分残留,肺大体及光镜病理观察与对照组相比均未产生明显变化。光镜病理观察可见海藻酸钙组的右肺底有局灶性肺泡破裂,肺泡间隔无炎性细胞浸润。卡波姆凝胶组的左肺底偶见局灶性肺不张。结论海藻酸钙和卡波姆凝胶肺内应用均具有较好的安全性,可作为含抗结核药物凝胶的良好基质。
Objective To evaluate the safety of calcium alginate and carbomer gel in bronchial bronchial interventional bronchus in rabbits, and provide the basis for the application of antimicrobial drugs in lungs. Methods Forty-five Japanese long-eared rabbits were randomly divided into control group, calcium alginate group and carbomer group, 15 rats in each group. The experiment was observed for 4 weeks. Body temperature, body weight, blood routine, T cell subsets and lung pathology were measured. Results There was no significant difference in body temperature, body weight, blood routine and peripheral T cell subsets between groups (P> 0.05). There was no significant difference in the percentages of CD3 +, CD4 + and CD8 + between the calcium alginate group and the carbomer group before intervention (P> 0.05). The alginate-calcium group and carbomer group showed no gel-like components in the left lung of the test animals, and no significant changes were observed in the lung and light microscope compared with the control group. Light microscopic examination showed that alginate group had a focal alveolar rupture in the right lung and no inflammatory cell infiltration in the alveolar space. Cavallum gel group of left lung occasionally seen atelectasis. Conclusion Both calcium alginate and carbopol gel have good safety in lung application and can be used as a good matrix for antituberculosis drug gels.
