系统研究了基于金纳米粒子的聚合酶链反应(PCR).通过Au-S键将5′端修饰-SH-(CH2)6-的引物连接在金纳米粒子表面,采用质粒pBluescriptSK作为模板,分别研究了引物浓度、退火温度和循环次数对PCR的影响.实验研究结果表明:在优化的条件下,将上游引物或下游引物连接在金纳米粒子表面以及将上游引物和下游引物分别连接在金纳米粒子表面,均能顺利进行PCR.基于金纳米粒子的PCR技术的建立,将均相体系中的PCR技术扩展到纳米粒子表面,不仅可以方便目标PCR产物的分离纯化,而且可以为制备连接在纳米粒子表面的ssDNA和纳米结构材料提供新的方法.
Polymerase chain reaction (PCR) based on gold nanoparticles was systematically studied, and the primers of 5’-end modified-SHH- (CH2) 6- were attached to the surface of gold nanoparticles by Au-S bond.
