目的:研究不同提取溶剂提取无梗五加根中总木脂素的最佳工艺。方法:采用紫外-可见分光光度法,以芝麻脂素为对照品,测定样品溶液在558 nm波长处的吸光度。采用正交试验法,以木脂素的含量为考察指标,对提取时间,提取次数及提取溶剂用量等影响因素进行研究。结果:对照品芝麻脂素取样量在10.6~53.0μg·ml-1范围内吸光度与浓度呈良好的线性关系(r=0.999 7)。平均回收率102.2%(RSD=1.6%,n=6)。以芝麻脂素计,无梗五加根中总木脂素含量为10.0 mg·g-1。最佳提取条件确定为无梗五加根药材粉末50%甲醇40倍量回流提取1次。结论:采用紫外分光光度法测定总木脂素含量方法简便、准确、可靠,最佳提取工艺可行。
Objective: To study the best extraction technology of total lignans from different roots and stems with different extraction solvents. Methods: The absorbance of the sample solution at 558 nm was measured by UV-Vis spectrophotometry with sesamin as reference substance. Orthogonal test was used to determine the content of lignans as the index, the extraction time, the number of extraction and the amount of solvent extraction factors were studied. Results: There was a good linear relationship between absorbance and concentration in the range of 10.6-53.0μg · ml-1 for the reference sesamellin sample (r = 0.999 7). The average recovery was 102.2% (RSD = 1.6%, n = 6). With sesamin, total lignans content in the roots of sessile stems was 10.0 mg · g-1. The optimum extraction conditions were determined as the non-stem Wujia root medicinal powder 50% methanol 40 times the amount of reflux extraction 1 times. Conclusion: The method of determination of total lignans by UV spectrophotometry is simple, accurate and reliable, and the best extraction process is feasible.
