目的探讨BMI-1mRNA和MMP-9mRNA在结肠癌细胞中的表达及其意义。方法采用半定量逆转录-聚合酶链反应(RT-PCR)方法检测结肠癌细胞和人正常结肠平滑肌细胞中BMI-1mRNA和MMP-9mRNA的表达,分析二者的关系。结果在结肠癌SW620和LoVo细胞株中,BMI-1mRNA的表达量明显高于结肠癌细胞株SW480细胞和人正常结肠平滑肌细胞(P<0.05);SW620和LoVo结肠癌细胞中MMP-9mRNA的表达亦明显高于SW480细胞和人正常结肠平滑肌细胞(P<0.05)。在结肠癌细胞中BMI-1mRNA的表达与MMP-9mRNA的表达呈正相关(r=0.966,P<0.05)。结论作为原癌基因,BMI-1可用作反映结肠癌细胞侵袭力的生物学指标。
Objective To investigate the expression of BMI-1 mRNA and MMP-9 mRNA in colon cancer cells and its significance. Methods The expression of BMI-1mRNA and MMP-9mRNA in colon cancer cells and human normal colon smooth muscle cells were detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). Results The expression of BMI-1 mRNA in colon cancer SW620 and LoVo cell lines was significantly higher than that in colon cancer cell line SW480 and human normal colonic smooth muscle cells (P <0.05). The expression of MMP-9 mRNA in SW620 and LoVo colon cancer cells Also significantly higher than SW480 cells and normal human colon smooth muscle cells (P <0.05). The expression of BMI-1 mRNA and MMP-9 mRNA in colon cancer cells were positively correlated (r = 0.966, P <0.05). Conclusion As a proto-oncogene, BMI-1 can be used as a biological indicator of colon cancer cell invasiveness.
