目的建立经口灌胃途经给予造模致敏原的挪威棕色(Brown Norway,BN)大鼠致敏动物模型。方法选用不同周龄(4周龄和8周龄)及不同性别(雄性和雌性)的BN大鼠,每天经口灌胃给予不同剂量(10.0、1.0、0.1mg)的造模致敏原—卵清蛋白(OVA),共35天。在第28和35天分别进行内眦静脉取血并分离血清,采用酶联免疫吸附测定(ELISA)方法检测血清中OVA特异性IgE(OVA sIgE)浓度。结果中剂量组雌性4周龄和8周龄BN大鼠血清中OVA sIgE浓度在第28和35天较阴性对照组差异有统计学意义(P<0.05);低剂量组雄性8周龄BN大鼠血清中OVA sIgE浓度在第28天较阴性对照组差异有统计学意义(P<0.05);各组雄性4周龄BN大鼠血清中OVA sIgE浓度在第28和35天均较阴性对照组差异无统计学意义。结论经口灌胃途径给予不同性别和周龄的BN大鼠不同剂量的OVA,雌性大鼠比雄性大鼠更敏感;周龄对敏感性无影响;1.0 mg的致敏剂量较适合。因此,选用雌性BN大鼠,每天经口灌胃给予1.0 mg OVA,28~35天即可建立比较理想的BN大鼠经口致敏动物模型。
OBJECTIVE: To establish an animal model of sensitization of Brown Norway (BN) rats by oral gavage. Methods BN rats of different weeks (4 weeks old and 8 weeks old) and different sexes (male and female) were given orally daily with different doses (10.0,1.0,0.1 mg) of model allergen - Ovalbumin (OVA) for a total of 35 days. On the 28th and 35th days, blood samples were taken from the internal canthal vein and the serum was separated. The serum concentration of OVA-specific IgE (OVA sIgE) was measured by enzyme-linked immunosorbent assay (ELISA). Results The serum concentration of OVA sIgE in middle-dose female BN rats at 4 weeks and 8 weeks was significantly lower than that of the negative control group on the 28th and 35th days (P <0.05). The low-dose male BN rats at 8 weeks The concentration of OVA sIgE in the serum of rats on the 28th day was significantly lower than that of the negative control (P <0.05). The concentrations of OVA sIgE in serum of the male 4-week-old BN rats were lower than those of the negative control group on the 28th and 35th days The difference was not statistically significant. Conclusion Oral administration of OVA at different doses and doses in BN rats of different sexes and weeks of age is more sensitive to female rats than that of male rats. There is no effect on the sensitivity of BN rats. The sensitizing dose of 1.0 mg is more suitable. Therefore, the female BN rats were selected, orally given by oral gavage 1.0 mg OVA, 28 to 35 days to establish a more ideal BN rat orally sensitized animal model.
