目的探讨直接注射移植人胚胎生殖(EG)细胞对大鼠心肌梗死的影响。方法40只SD大鼠经缝扎左冠状动脉前降支,建立急性心肌梗死(AMI)模型,分干细胞移植组和对照组。取5~10周人胚胎生殖腺嵴,组织块体外培养,生物学鉴定证实为人EG细胞,将其直接注入大鼠急性心肌梗死区边缘。移植后1 d,1、2、4周处死大鼠,采用免疫组化法观察心肌形成转录因子Nkx 2.5在移植细胞的表达。结果移植组鼠抗人细胞核抗体MAB1281检测阳性,心肌形成转录因子Nkx2.5在移植细胞阳性表达。结论人EG细胞直接注入移植大鼠心肌梗死处,细胞能存活并呈现向心肌细胞分化的表型。
Objective To investigate the effect of direct injection of human embryonic germ (EG) cells on myocardial infarction in rats. Methods 40 SD rats were established by anterior descending branch of left anterior descending coronary artery and established acute myocardial infarction (AMI) model, divided into stem cell transplantation group and control group. Take 5 to 10 weeks of human embryonic gonadal cristae, tissue culture in vitro, biological identification confirmed as human EG cells, injected directly into the edge of acute myocardial infarction area. At 1, 2, and 4 weeks after transplantation, rats were sacrificed and the expression of myocardial transcription factor Nkx 2.5 in the transplanted cells was observed by immunohistochemistry. Results MAB1281 anti-human nuclear antibody MAB1281 in the transplanted group was positive, and the expression of myocardial transcription factor Nkx2.5 in the transplanted cells was positive. CONCLUSION: Human EG cells are directly injected into myocardial infarction of transplanted rats, and the cells can survive and present the phenotype differentiated into cardiomyocytes.
