目的:探讨荔枝核皂苷对乳腺增生大鼠雌激素受体ERα、ERβ及ERK通路的影响。方法:取未孕雌性SD大鼠,除留正常对照组8只外,其他动物均进行造模,成模动物随机分为模型组、荔枝核皂苷低剂量组(0.1 g/kg)、荔枝核皂苷高剂量组(0.2 g/kg)和他莫昔芬阳性对照组(4 mg/kg),每组8只。以上各组分别灌胃给药30 d后,采用免疫组化方法检测乳腺组织ERα、ERβ、细胞外信号调节激酶(ERK)及血管内皮生长因子(VEGF)的表达。结果:与模型组比较,荔枝核皂苷高剂量组大鼠乳腺组织ERα、ERK、VEGF的表达显著降低(P<0.05);低剂量组大鼠乳腺组织ERα、ERK的表达显著降低(P<0.05)。结论:荔枝核皂苷可降低乳腺增生大鼠乳腺组织中ERα、ERK、VEGF的表达。
Objective: To investigate the effects of lychee nuclear saponins on estrogen receptor ERα, ERβ and ERK pathways in rats with hyperplasia of mammary glands. Methods: The non-pregnant female Sprague Dawley rats were divided into model group, low-dose group (0.1 g / kg), litchi High dose saponin group (0.2 g / kg) and tamoxifen positive control group (4 mg / kg), each group of eight. The above groups were administered intragastrically for 30 days. The expression of ERα, ERβ, ERK and VEGF in breast tissues were detected by immunohistochemistry. Results: Compared with the model group, the expressions of ERα, ERK and VEGF were significantly decreased (P <0.05) in the livers of the high-dose litchi nucleus group; the expression of ERα and ERK in the low- ). Conclusion: Lychee nuclear saponin can reduce the expression of ERα, ERK and VEGF in mammary gland tissue of rats with hyperplasia of mammary gland.
