中脑多巴胺(DA)能神经元的发育成熟过程主要有两条转录因子信号通路参与一条是孤儿核受体(Nurr1)-酪氨酸羟化酶(TH)通路调控DA神经递质合成;另一条通路需要同源域蛋白Lmx1b和En的参与,Lmx1b驱动Pitx3控制中脑DA能神经元的成熟、存活,En1、En2(engrailed1、2)则控制其存活。中脑DA能神经元的正常发育是多种信号分子通路共同调控,DA能神经元的分化成熟过程是神经发育学研究的重要分支,对于阐明相关神经精神疾病的病理机制、寻找治疗方案,尤其是对帕金森病的发病和治疗意义重大。
The process of development and maturation of dopaminergic neurons in the midbrain mainly involves two transcription factor signaling pathways involved in the synthesis of DA neurotransmitter mediated by the Nurr1-tyrosine hydroxylase (TH) pathway. One pathway requires the participation of the cognate domain proteins Lmx1b and En. Lmx1b drives Pitx3 to control the maturation and survival of DA neurons in the midbrain, while En1 and En2 (engrailed1,2) control their survival. The normal development of midbrain DA neurons is regulated by a variety of signaling pathways. The process of differentiation and maturation of DA neurons is an important branch of neurodevelopment research. To clarify the pathological mechanism of related neuropsychiatric disorders and find the treatment plan, especially It is significant for the pathogenesis and treatment of Parkinson’s disease.
