灯盏花素对脑缺血再灌注大鼠海马Akt表达的影响

目的探讨灯盏花素对脑缺血再灌注大鼠海马Akt/(PKB)表达的影响。方法采用Zea-Longa法建立脑缺血再灌注动物模型。72只SD大鼠随机分为:假手术组(Sham,8只)、灯盏花素治疗组(Br,32只)、缺血再灌注组(I/R,32只)。Br治疗组和I/R组依据缺血后再灌注的不同时间点再细分为3、12,24、48h4个小组。应用免疫组织化学方法和蛋白印迹法检测各组大鼠海马Akt蛋白的表达。结果与假手术组相比,在缺血再灌注3h时,大鼠海马Akt蛋白的阳性表达已明显降低,在24h时表达量最低,随后表达开始上升(P<0.05);与相同时间点的缺血再灌注组大鼠比较,3h时,Br治疗组的Akt蛋白阳性表达未见明显降低(P>0.05),其余各时间点则明显降低(P<0.05);蛋白印迹法也得到了相同的结论。结论灯盏花素很可能通过上调Akt蛋白的表达改善脑缺血再灌注损伤。 Objective To investigate the effect of breviscapine on the expression of Akt / (PKB) in hippocampus of cerebral ischemia-reperfusion rats. Methods The animal model of cerebral ischemia-reperfusion was established by Zea-Longa method. Seventy-two Sprague-Dawley rats were randomly divided into sham-operation group (Sham, 8), breviscapine treatment group (32) and ischemia / reperfusion group (32 rats). Br treatment group and I / R group were further subdivided into 3, 12, 24, 48h4 groups according to different time points after ischemia and reperfusion. The expression of Akt protein in hippocampus of rats in each group was detected by immunohistochemistry and Western blotting. Results Compared with the sham-operation group, the expression of Akt protein in hippocampus of rats was significantly decreased at 3h after ischemia / reperfusion and lowest at 24h, then the expression began to increase (P <0.05) At 3h, the expression of Akt protein in Br group was not significantly reduced (P> 0.05), but significantly decreased in other groups (P <0.05); Western blotting was also the same Conclusion. Conclusion Breviscapine may improve cerebral ischemia-reperfusion injury by up-regulating the expression of Akt protein.
