目的分析吉林省2001~2014年间麻疹病毒血凝素(hemagglutinin,H)蛋白基因的亲缘关系。方法选取35株吉林省麻疹病毒株作为代表株,采用RT-PCR法获得麻疹病毒H蛋白核酸分段扩增产物,将测得的序列用DNASTAR软件进行拼接,并用Mega 4.0和Bioedit软件对序列进行亲缘关系和同源性分析。结果 35株麻疹病毒代表株均属于H1基因型H1a基因亚型,且明显处于两个小分支上。35株麻疹病毒株H蛋白基因同源性变化为:株间核苷酸同源性为98.2%~99.8%,株间氨基酸同源性为95.2%~99.8%;与H1a中国代表株(China93-4/H1a)相比,核苷酸同源性为98.8%~99.6%,氨基酸同源性为96.7%~98.8%;与中国疫苗株(Shanghai-191)相比,核苷酸同源性为94.5%~95.2%,氨基酸同源性为84.7%~86.7%。结论吉林省2001~2014年间,麻疹病毒H蛋白与中国代表株(China93-4/H1a)的亲缘关系呈逐年渐远的态势发展,与中国H1a代表株和中国疫苗株相比,核苷酸和氨基酸同源性均呈逐年下降的趋势,提示应对吉林省麻疹病毒H蛋白进行持续监测和分析。
Objective To analyze the genetic relationship of hemagglutinin (H) gene in Jilin Province from 2001 to 2014. Methods 35 strains of measles virus in Jilin Province were selected as representative strains. The amplified products of measles virus H protein nucleic acids were obtained by RT-PCR. The sequences were stitched with DNASTAR software and sequenced by Mega 4.0 and Bioedit software Phylogenetic relationship and homology analysis. Results 35 strains of measles virus belong to the H1 genotype H1a subtype, and apparently in two small branches. The H gene homology of 35 measles isolates varied from 98.2% to 99.8% for nucleotide and 95.2% to 99.8% for isolates, 4 / H1a), nucleotide homology was 98.8% -99.6% and amino acid homology was 96.7% -98.8%. Compared with the Chinese vaccine strain Shanghai-191, the nucleotide homology was 94.5% ~ 95.2%, amino acid homology was 84.7% ~ 86.7%. Conclusion The genetic relationship between H-protein of measles virus and Chinese representative strain (China93-4 / H1a) showed a gradual declining trend from 2001 to 2014 in Jilin Province. Compared with the Chinese H1a strain and the Chinese vaccine strain, nucleotide and Amino acid homology showed a trend of declining year by year, suggesting that the response to Jilin Province, measles virus H protein for continuous monitoring and analysis.
