目的:探讨MDM2、p53和p21WAF1蛋白在不同级别脑胶质瘤组织中的表达及其在胶质瘤发生、发展中的生物学意义。方法:应用免疫组化SP法检测Ⅱ级和Ⅳ级胶质瘤组织中MDM2、p53和p21WAF1蛋白的表达,并分析其与胶质瘤组织学分级之间的关系。结果:Ⅱ级和Ⅳ级胶质瘤中,MDM2蛋白的阳性表达率分别为24.00%(6/25)和56.52%(13/23),χ2=5.298,P=0.021,p53蛋白的阳性表达率分别为28.00%(7/25)和60.87%(14/23),χ2=5.259,P=0.022,两者阳性表达率均随胶质瘤恶性程度的增加而升高,Spearman等级相关分析显示,MDM2、p53的表达与胶质瘤分级均呈正相关,P<0.05;p21WAF1的阳性表达率分别为76.00%(19/25)和39.13%(9/23),χ2=6.700,P=0.010,其阳性表达率随胶质瘤恶性程度的增加而降低,Spearman等级相关分析显示,p21WAF1的表达与胶质瘤分级呈负相关,r=-0.425,P=0.003。结论:MDM2蛋白和突变型p53蛋白的过表达以及p21WAF1蛋白的低表达,可促进胶质瘤的发生、发展。
Objective: To investigate the expression of MDM2, p53 and p21WAF1 protein in different grades of glioma tissue and its biological significance in the development and progression of glioma. Methods: The expressions of MDM2, p53 and p21WAF1 protein in grade Ⅱ and Ⅳ gliomas were detected by immunohistochemical SP method, and their relationship with histological grading of glioma was analyzed. Results: The positive expression rates of MDM2 protein in grade Ⅱ and grade Ⅳ gliomas were 24.00% (6/25) and 56.52% (13/23) respectively, χ2 = 5.298, P = 0.021. The positive rate of p53 protein Respectively. The positive expression rates of them were 28.00% (7/25) and 60.87% (14/23) respectively, χ2 = 5.259, P = 0.022. The Spearman rank correlation analysis showed that the positive expression rates of both of them were increased with the malignant degree of glioma, The positive rates of p21WAF1 were 76.00% (19/25) and 39.13% (9/23), respectively, χ2 = 6.700 and P = 0.010, respectively The positive rate of glioma decreased with the increase of malignant degree. Spearman rank correlation analysis showed that the expression of p21WAF1 was negatively correlated with the grade of glioma (r = -0.425, P = 0.003). Conclusion: Overexpression of MDM2 protein and mutant p53 protein and low expression of p21WAF1 protein may promote the development of glioma.
