目的探讨Wnt7a/PCP信号通路在肾衰营养胶囊改善CRF大鼠骨骼肌萎缩中的作用及机制。方法将大鼠随机分为正常组、模型组、中药组和开同组,采用切除5/6肾并予4%酪蛋白饲料喂养制作CRF营养不良大鼠模型。采用免疫组化法检测TA中Myogenin和Myo D的表达;采用Western blot检测Wnt7a/PCP信号通路中的蛋白表达。结果肾衰营养胶囊增加CRF大鼠体重和骨骼肌质量,增加TA中Myogenin和Myo D蛋白的表达,促进骨骼肌组织Wnt7a/PCP信号通路中蛋白表达上调。结论肾衰营养胶囊具有改善CRF模型大鼠骨骼肌萎缩的作用,其机制可能与上调骨骼肌Wnt7a/PCP信号通路、促进MSC增殖分化相关。
Objective To investigate the role and mechanism of Wnt7a / PCP signaling pathway in improving renal atrophy in skeletal muscle of CRF rats. Methods The rats were randomly divided into normal group, model group, traditional Chinese medicine group and control group. CRF malnutrition rat model was made by cutting 5/6 kidney and feeding 4% casein diet. The expression of Myogenin and Myo D in TA was detected by immunohistochemistry. The protein expression in Wnt7a / PCP signal pathway was detected by Western blot. Results Shenfubun capsule increased the body weight and skeletal muscle mass of CRF rats, increased the expression of Myogenin and Myo D in TA, and promoted the up-regulation of Wnt7a / PCP signaling in skeletal muscle. Conclusion Shenshuai Nourishing Capsule can ameliorate skeletal muscle atrophy in rats with CRF. The mechanism may be related to up-regulation of Wnt7a / PCP signaling pathway and promotion of proliferation and differentiation of MSCs.
