目的 :观察冠心病 (CHD)患者外周血内皮祖细胞 (endothelialprogenitorcells,EPC)数量和功能的改变。方法 :选择CHD患者和非CHD患者各 2 0例 ,用密度梯度离心法从外周血获取单个核细胞 ,将其接种在人纤维连接蛋白包被培养板 ,培养 7d后贴壁细胞进行细胞化学分析。激光共聚焦显微镜鉴定FITC标记荆豆凝血素Ⅰ和DiI 标记的乙酰化低密度脂蛋白双染色阳性细胞为正在分化的EPC。采用二苯基四氮唑嗅盐比色法、改良的Boyden小室和黏附能力测定实验观察EPC的增殖能力、迁移能力和黏附能力。 结果 :CHD患者外周血EPC数量明显减少 [(31.8± 7.7)∶(5 9.5± 10 .6 )EPC/视野 (× 2 0 0 ) ,P <0 .0 5 ],且CHD患者外周血EPC的黏附能力、迁移能力和增殖能力也明显受损。结论 :CHD患者外周血EPC数量减少、功能减退。
Objective: To observe the quantity and function of endothelial progenitor cells (EPCs) in patients with coronary heart disease (CHD). METHODS: Twenty patients with CHD and non-CHD were selected. Mononuclear cells were obtained from peripheral blood by density gradient centrifugation. The cells were inoculated on human fibronectin coated plates and cultured for 7 days. The adherent cells were cultured for cytochemical analysis . Laser confocal microscopy identified FITC-labeled daidzein-Ⅰ and DiI-labeled acetylated low-density lipoprotein-positive double-stained cells were differentiated EPCs. The proliferation, migration and adhesion of EPC were observed by diphenyltetrazolium chloride salt assay, modified Boyden chamber and adhesion assay. Results: The number of EPC in peripheral blood of patients with CHD was significantly decreased ([(31.8 ± 7.7) vs (9.5 ± 10.6) EPCs / field of vision (× 200), P <0.05] Adhesion, migration and proliferation were also significantly impaired. Conclusion: The number of EPC in peripheral blood of patients with CHD is decreased and the function is diminished.
