目的:探讨清化颗粒调节db/db糖尿病小鼠肠道钠-葡萄糖协同转运蛋白1(SGLT1)的表达促进小鼠胰高血糖素样肽-1(GLP-1)的分泌。方法:取6只db/m小鼠为空白对照组,24只db/db糖尿病小鼠随机分为模型对照组,清化颗粒低、中、高剂量组,每组6只,连续灌胃给药4周,观察小鼠空腹血糖水平变化,ELISA法分析小鼠GLP-1水平、q RT-PCR、Western blot和免疫组化技术分别检测小鼠肠道组织SGLT1基因和蛋白的表达水平。结果:与模型对照组比较,清化颗粒下调db/db糖尿病小鼠的空腹血糖水平,上调血清GLP-1水平(P<0.01),并具有剂量依赖性。清化颗粒显著上调肠道组织中SGLT1基因和蛋白的表达(P<0.01)。结论:清化颗粒能够降低db/db糖尿病小鼠的血糖水平,促进GLP-1分泌,其作用机制可能与上调肠道组织中SGLT1的表达有关。
AIM: To investigate the effect of Qinghua Granule on the expression of intestinal sodium-glucose cotransporter 1 (SGLT1) in db / db diabetic mice and promote the secretion of mouse glucagon-like peptide-1 (GLP-1). Methods: 6 db / m mice were taken as blank control group. Twenty-four db / db diabetic mice were randomly divided into model control group, Qinghua Granule low, medium and high dose group, 6 rats in each group. The changes of fasting blood glucose level in mice were observed after 4 weeks. The level of GLP-1 in mice was analyzed by ELISA. The expression of SGLT1 gene and protein in intestinal tract were detected by q RT-PCR, Western blot and immunohistochemistry. Results: Compared with the model control group, Qinghua Granule reduced fasting blood glucose level and raised serum GLP-1 level in db / db diabetic mice (P <0.01) in a dose-dependent manner. Qinghua Granule could up-regulate the expression of SGLT1 gene and protein in intestinal tissue (P <0.01). Conclusion: Qinghua Granule can reduce the blood glucose level and promote the secretion of GLP-1 in db / db diabetic mice. Its mechanism may be related to the up-regulation of SGLT1 expression in intestinal tract.
