目的:建立中药玄参中哈巴俄苷和肉桂酸的高效液相色谱含量测定方法,提出特征性有效成分哈巴俄苷的含量限度建议。方法:Ultrasphere ODS 分析柱 (250 mm 4.6 mm,5 mm),流动相为乙腈和1% 醋酸水溶液,线性梯度洗脱(20:8050:50) (20 min),流速为 1 mLmin-1,室温,检测波长为278 nm。结果:哈巴俄苷与肉桂酸回收率分别为97.13% 和99.38%,RSD分别为0.80% 和0.51%,全国20份商品饮片和8份不同产地完整药材中哈巴俄苷和肉桂酸的含量分别为0.041~0.244% 和0.012~0.068%。结论:本文方法快速、简便、准确,可用于玄参的质量控制。建议规定玄参中哈巴俄苷含量应不低于0.05%。
Objective: To establish a HPLC method for the determination of harpagoside and cinnamic acid in Scrophularia scrophulariae, and to propose a limit for the content of harpagoside, a characteristic active ingredient. Method: Ultrasphere ODS column (250 mm 4.6 mm, 5 mm), mobile phase of acetonitrile and 1% aqueous acetic acid, linear gradient elution (20:8050:50) (20 min), flow rate 1 mLmin-1, room temperature The detection wavelength is 278 nm. RESULTS: The recoveries of harpagoside and cinnamic acid were 97.13% and 99.38%, RSDs were 0.80% and 0.51%, respectively. The content of harpagoside and cinnamic acid in 20 commercial decoction pieces and 8 complete production materials from different regions were 0.041~0.244% and 0.012~0.068%. Conclusion: This method is rapid, simple, accurate and can be used for quality control of Radix Scrophulariae. It is suggested that the content of harpagoside in Radix Scrophulariae should not be lower than 0.05%.
