目的探讨血小板衍生生长因子(PDGF)单独应用及与其受体抑制剂STI 571联合应用对人脐带间充质干细胞(MSCs)移行的影响,以探讨PDGF及STI 571对MSCs变化的影响,为评估MSCs的临床应用提供依据。方法采用细胞划痕的方法观察不同时间点MSCs的迁移情况。结果 PDGF单独应用及与STI 571联合应用对MSCs迁移的影响,单用PDGF组(EG1)较对照组迁移快;PDGF及STI 571二者联合应用组(EG2)迁移较单用PDGF组慢。结论①PDGF可以促进MSCs迁移。②联合应用PDGF与STI 571可以抑制MSCs迁移。
Objective To investigate the effect of platelet-derived growth factor (PDGF) alone and in combination with its receptor inhibitor STI 571 on the migration of human umbilical cord mesenchymal stem cells (MSCs) to explore the effect of PDGF and STI 571 on the changes of MSCs. Provide the basis for clinical application. Methods The cell scratch was used to observe the migration of MSCs at different time points. Results The effect of PDGF alone or in combination with STI 571 on MSCs migration was faster in PDGF group than that in control group. The combined application of PDGF and STI 571 group (EG2) was slower than PDGF group alone. Conclusion ① PDGF can promote the migration of MSCs. ② Combination of PDGF and STI 571 can inhibit MSCs migration.
