目的:探讨脂氧合酶抑制剂NDGA在体外对结肠癌HT-29细胞株生长抑制、凋亡影响。方法: 应用MTT法绘制生长曲线;倒置相差显微镜观察细胞形态变化;扫描电镜观察细胞的超微结构变化, 检测凋亡小体;流式细胞仪检测细胞凋亡、细胞周期变化。结果:不同浓度NDGA处理HT-29结肠癌细胞,随着药物浓度加大,细胞形态变圆,体积缩小,从瓶壁上逐步脱落,肿瘤细胞凋亡率逐步上升,癌细胞阻滞于G0/G1期。应用扫描电镜可见凋亡小体形成。结论:NDGA对结肠癌HT-29细胞具有抑制生长、诱导凋亡作用,凋亡率随药物浓度加大逐步升高。具有剂量依赖效应。
Objective: To investigate the effect of lipoxygenase inhibitor NDGA on the growth inhibition and apoptosis of colon cancer HT-29 cells in vitro. Methods: The growth curve was drawn by MTT method. Morphological changes were observed by inverted phase contrast microscope. The ultrastructural changes of the cells were observed by scanning electron microscope. The apoptotic bodies were detected by flow cytometry. The apoptosis and cell cycle were detected by flow cytometry. Results: After treated with different concentrations of NDGA, HT-29 colon cancer cells showed a round shape with a decreasing volume and a gradual detachment from the wall of the bottle. The apoptosis rate of tumor cells gradually increased and the number of cancer cells arrested in the G0 / G1 phase. The appearance of apoptotic bodies can be seen by scanning electron microscopy. Conclusion: NDGA can inhibit the growth of colon cancer HT-29 cells and induce apoptosis. The apoptosis rate increases with the increase of drug concentration. A dose-dependent effect.
