目的 :研究衰老大鼠肺部感染时血浆血管加压素(Argininevasopressin ,AVP)浓度的变化及其与心脏胶原蛋白之间的关系 .方法 :雄性Sprague Dawley (SD)大鼠经scD 半乳糖人工老化后随机分为 3组 .①模型组 :气管穿刺注入 0 .2mL肺炎球菌菌液诱发肺炎 ,并未予治疗 .②抗生素组 :气管穿刺注入 0 .2mL肺炎球菌菌液诱发肺炎并于第 3日开始静脉注射青霉素钠 5× 1 0 5U/(kg·d) ,连续注射 1 3d .③对照组 :气管穿刺注入 0 .2mL生理盐水 .于注菌后第 1 5日各组均开胸取心脏并留取血浆 ,分离左右心室 ,取左室前壁匀浆 ,光学比色法检测羟脯氨酸浓度并计算胶原蛋白浓度 ;放射免疫法检测血浆AVP浓度 .结果 :①模型组血浆AVP浓度[(0 .78± 0 .1 7) μg/L]高于对照组 [(0 .5 9± 0 .0 8) μg/L]和抗生素组 [(0 .5 5± 0 .0 9) μg/L],均具有非常显著性差异 (P <0 .0 1 ) .对照组和抗生素组血浆AVP浓度比较无显著性差异 (P>0 .0 5 ) .②模型组心脏胶原蛋白浓度 [(6 .5 4± 0 .91 )mg/g]高于对照组 [(4 .77± 0 .98)mg/g]和抗生素组 [(5 .2 2± 0 .83)mg/g],均具有统计学意义 (P <0 .0 1 ) .对照组和抗生素组心脏胶原蛋白浓度比较无显著性差异 (P >0 .0 5 ) .③肺部感染时衰老大鼠心脏胶原蛋白浓度随血浆AVP浓度升高而增?
Objective: To investigate the changes of plasma concentration of arginine vasopressin (AVP) and the relationship with the level of cardiac collagen in the lungs of aging rats.Methods: Male Sprague Dawley (SD) rats were induced to undergo artificial aging by scD galactose And then randomly divided into 3 groups: ① model group: 0.2 ml pneumococcal fluid was injected into the lung by pneumotracheal intratracheal instillation of pneumonia, and no treatment was given.② Antibiotic group: 0.2 ml pneumococcal fluid was injected into bronchial tube to induce pneumonia and on the 3rd day Into the penicillin sodium intravenous injection of 5 × 105 U / (kg · d), continuous injection of 1 3d. ③ control group: tracheal injection of 0. 2mL saline. Plasma samples were collected and the left and right ventricles were separated and homogenated in the anterior wall of the left ventricle. The concentration of hydroxyproline in the left ventricle was measured by optical colorimetry and the concentration of collagen was calculated. The plasma AVP concentration was measured by radioimmunoassay.Results: (0.78 ± 0.17) μg / L] were higher than those in the control group [(0.59 ± 0.08) μg / L] and the antibiotic group [(0.55 ± 0.09) μg / L], there was a significant difference (P <0.01). There was no significant difference in plasma AVP concentrations between the control group and the antibiotic group (P> 0 .⑤5) .②The concentration of cardiac collagen in the model group [(6.64 ± 0.91) mg / g] was higher than that in the control group [(4.77 ± 0.98) mg / g] and the antibiotic group 5 2.2 ± 0.83 mg / g], all of which were statistically significant (P <0.01). There was no significant difference in cardiac collagen concentration between the control group and the antibiotic group (P> 0.05) .③The concentration of collagen in the heart of aging rats increased with the increase of plasma AVP concentration in pulmonary infection.
