建立HPLC测定虫草菌粉中腺苷含量的分析方法。采用色谱柱为Kromasil C18(150mm×4.6mm,5μm),乙腈∶水=5∶95为流动相,流速为1.0mL/min,检测波长为260nm,柱温为室温,进样量为5μL。结果显示每个样品分析时间约为12min,保留时间(tR)约9.0—9.6min,腺苷的线性方程为y=11.659x-4.215,线性范围12.5—62.5ng,r=0.9996,回收率为99.00%,RSD为1.11%。该方法灵敏度高、选择性好、样品前处理方法简单快速,回收率符合实验要求,适合于虫草菌粉中腺苷的含量分析。
Establishment of HPLC method for the determination of adenosine in Cordyceps powder. The column was Kromasil C18 (150 mm × 4.6 mm, 5 μm). Acetonitrile: water = 5:95 was used as the mobile phase at a flow rate of 1.0 mL / min. The detection wavelength was 260 nm. The column temperature was room temperature and the injection volume was 5 μL. The results showed that the analysis time of each sample was about 12min, the retention time (tR) was about 9.0-9.6min, the linear equation of adenosine was y = 11.659x-4.215, the linear range was 12.5-62.5ng, r = 0.9996, the recovery was 99.00 %, RSD is 1.11%. The method has high sensitivity, good selectivity, simple and rapid sample preparation method, and the recovery meets the experimental requirements and is suitable for the content analysis of adenosine in Cordyceps powder.
