冬枣提取物对S_(180)荷瘤小鼠的免疫及抗氧化作用(英文)

目的观察冬枣提取物(DJE)对S180荷瘤小鼠的免疫及抗氧化作用。方法取昆明种小鼠50只,先设正常对照组10只,其余小鼠左腋皮下接种瘤液0.2 mL,24 h后随机分为肿瘤对照组、冬枣乙醇提取物(EED)组、冬枣乙酸乙酯萃取物(EAED)组,环磷酰胺(CTX)组。正常对照组、肿瘤对照组用生理盐水灌胃,每天1次;EED组、EAED组分别给予EED 100 mg/(kg.d)、EAED 100 mg/(kg.d)灌胃;CTX组给予CTX 20 mg/kg,第2天起隔日腹腔注射1次。实验连续10 d。末次给药24 h后,将小鼠称质量。眼眶取血,测定各组丙二醛(MDA)含量及超氧化物歧化酶(SOD)活力,采用双抗体夹心ELISA法测定各组血清白细胞介素2浓度。剥取肿瘤、胸腺、脾脏,计算抑瘤率、胸腺指数、脾指数。结果DJE可明显抑制S180荷瘤小鼠瘤体生长,肿瘤抑制率最高达到24.8%;DJE干预组胸腺指数、脾指数与肿瘤对照组比较均无明显差异(P>0.05),但均较CTX组显著增高(F=3.912、8.924,q=2.502~4.654;P<0.05);DJE干预组小鼠血清中白细胞介素-2含量及SOD活力均较肿瘤对照组显著增加(F=2.932、41.299,q=2.839~12.222,P<0.05),MDA含量则明显低于肿瘤对照组(F=12.917,q=5.812、3.196,P<0.05)。结论DJE具有抑制S180实体瘤生长,提高荷瘤小鼠免疫功能以及增强抗氧化防御系统酶活性的作用。 Objective To observe the immune and antioxidation effects of winter jujube extract (DJE) on S180 tumor-bearing mice. Methods A total of 50 Kunming mice were selected. Ten normal control groups were set first. The remaining mice were inoculated subcutaneously with 0.2 mL of tumor fluid. After 24 hours, they were randomly divided into tumor control group, ethanol extract of winter date (EED) group, and winter date acetic acid. Ethyl acetate extract (EAED) group, cyclophosphamide (CTX) group. Normal control group and tumor control group were intragastrically administered with normal saline once a day; EED group and EAED group were given EED 100 mg/(kg.d) and EAED 100 mg/(kg.d) gavage; CTX group was given CTX. 20 mg/kg, 1 intraperitoneal injection every other day from the second day. The experiment was continued for 10 days. The mice were weighed 24 hours after the last dose. The blood was taken from the eyelids, and the malondialdehyde (MDA) content and superoxide dismutase (SOD) activity were measured. The serum interleukin 2 concentration was measured by double antibody sandwich ELISA. The tumors, thymus, and spleen were removed, and the tumor inhibition rate, thymus index, and spleen index were calculated. Results DJE significantly inhibited the tumor growth in S180 tumor-bearing mice, with the tumor inhibition rate being up to 24.8%. There was no significant difference in thymus index and spleen index between the DJE intervention group and the tumor control group (P>0.05), but they were all higher than those of the CTX group. Significantly increased (F=3.912, 8.924, q=2.502 to 4.654; P<0.05); Serum levels of interleukin-2 and SOD in the DJE intervention group were significantly higher than those in the tumor control group (F=2.932, 41.299, q = 2.839 ~ 12.222, P <0.05), MDA content was significantly lower than the tumor control group (F = 12.917, q = 5.812, 3.196, P <0.05). Conclusion DJE can inhibit the growth of S180 solid tumor, increase the immune function of tumor-bearing mice and enhance the activity of antioxidant defense system.
