目的 :观察从柴胡中提取的单体成分柴胡皂甙d诱导白血病细胞株K562细胞凋亡及对其基因表达的影响。方法 :分别用 5μg/ml、7.5μg/ml、1 0 μg/ml浓度的SSd作用于K562细胞株 ,于药物作用后不同时间段 (1 2 ,2 4 ,36 ,48,60h) ,荧光染色法观察基凋亡状况、间接免疫荧光法测Bcl- 2表达情况。结果 :荧光染色结果显示 :K562在 1 0 μg/ml处理 2 4h后 ,开始出现凋亡小体 ,48h后出现大量死细胞。间接免疫荧光法测Bcl- 2表达 ;经SSd作用后K562细胞Bcl- 2基因表达下降
Objective : To observe the apoptosis of leukemic cell line K562 cells induced by saikosaponin d, a monomeric component extracted from Radix Bupleuri, and its effect on gene expression. METHODS: K562 cell lines were treated with 5 μg/ml, 7.5 μg/ml, and 10 μg/ml SSd, respectively, at different time periods (1 2, 24, 36, 48, and 60 h) after the drug treatment. Observed the apoptotic status of the method, indirect immunofluorescence method to measure the expression of Bcl-2. RESULTS : The results of fluorescent staining showed that apoptotic bodies began to appear after K562 cells were treated at 10 μg/ml for 24 hours. A large number of dead cells appeared after 48 hours. Bcl-2 expression was measured by indirect immunofluorescence; Bcl-2 gene expression was decreased in K562 cells treated with SSd.
