敦煌固本方对运动负荷大鼠股四头肌相关酶活性及心肌保护作用的研究

目的:探讨敦煌固本方对运动负荷大鼠股四头肌相关酶活性及对心肌的保护作用。方法:选用健康雄性大鼠60只,饲养1周后。随机分为运动负荷组和非负荷组,每组30只,运动负荷组再随机分为空动组和药动组,每组15只;非运动负荷组再随机分为空静组和药静组,每组15只。苦味酸标记后,药静组和药动组等体积灌胃注射敦煌固本方实验液0.4 mL,空静组和空动组注射等体积0.9%NaCl溶液,连续8周。训练方式为递增负荷游泳训练,持续训练8周。8周后断头处死大鼠,分别取心肌组织样品和股四头肌组织样品,采用比色法检测有关生理生化指标活性。结果:空静组与药动组琥珀酸脱氢酶(SDH)活力比较,差异有显著性意义(P<0.05),药静组与空静组SDH活力比较,差异有非常显著性意义(P<0.01)。药动组与药静组比较苹果酸脱氢酶(MDH)活力、乳酸脱氢酶(LDH)活力比较,差异均有显著性意义(P<0.05)。药动组与药静组丙二醛(MDA)、乳酸(LD)含量和总超氧化物歧化酶(SOD)活力比较,差异均有显著性意义(P<0.05)。结论:敦煌固本方能显著改善运动负荷造成的心肌、骨骼肌缺血缺氧状况。 Objective: To investigate the effect of Dunhuang Gushen Prescription on quail -related enzyme activity and myocardial protection in rats with exercise stress. Methods: Sixty healthy male rats were selected and fed for one week. The rats were randomly divided into exercise load group and non-load group, 30 rats in each group. The exercise load group was randomly divided into empty exercise group and pharmacokinetic group, 15 rats in each group.Non-exercise stress group was randomly divided into static group and drug- Group, 15 in each group. After picric acid labeling, drug-static group and pharmacokinetic group were injected intragastrically with 0.4 mL of Dunhuang Guben prescription liquid, and empty-group and empty-group were injected with 0.9% NaCl solution for 8 weeks. The training mode is incremental load swimming training, continuing training for 8 weeks. After 8 weeks, the rats were killed by decapitation, and the myocardial tissue samples and quadriceps femoris samples were taken respectively. The physiological and biochemical indexes were detected by colorimetric assay. Results: There was significant difference in the activity of succinate dehydrogenase (SDH) between the static group and the pharmacokinetic group (P <0.05). The difference of SDH activity between the static group and the empty group was significant (P <0.01). Pharmacokinetic group and drug static group malate dehydrogenase (MDH) activity, lactate dehydrogenase (LDH) activity comparison, the difference was significant (P <0.05). The differences of MDA, LD and SOD between YM and YJ groups were significant (P <0.05). Conclusion: Dunhuang Guben can significantly improve myocardial ischemia and hypoxia caused by exercise load.
