目的观察正常及急性淋巴细胞白血病骨髓基质细胞微环境对Jurkat细胞生物学特性的影响。方法收集与正常及急性淋巴细胞白血病骨髓基质细胞微环境共培养10d的Jurkat细胞,观测Jurkat细胞增殖、细胞周期分布、凋亡、分化及DNR摄药量的改变。结果白血病骨髓基质细胞微环境对Jurkat细胞黏附、趋化及降低摄药量的作用强于正常骨髓基质细胞微环境;抑制增殖及促分化作用弱于正常骨髓基质细胞微环境。白血病骨髓基质细胞微环境抑制Jurkat细胞凋亡,而正常骨髓基质细胞微环境促进其凋亡。结论正常骨髓源基质细胞微环境较白血病源骨髓基质细胞微环境对Jurkat细胞具有更强的促进凋亡、分化及抑制增殖的作用,增加Jurkat细胞对DNR的敏感性。
Objective To observe the effects of bone marrow stromal cell microenvironment on the biological characteristics of Jurkat cells in normal and acute lymphoblastic leukemia patients. Methods Jurkat cells co-cultured with normal and acute lymphoblastic leukemia bone marrow stromal cells (DMEM) for 10 days were collected. Proliferation, cell cycle distribution, apoptosis, differentiation and DNR uptake were observed in Jurkat cells. Results The leukemic bone marrow stromal cells microenvironment had stronger effects on the Jurkat cells adhesion, chemotaxis and drug intake than the normal bone marrow stromal cells microenvironment; the inhibition of proliferation and differentiation was weaker than that of normal bone marrow stromal cells. Leukemia bone marrow stromal cell microenvironment inhibits Jurkat cell apoptosis, whereas normal bone marrow stromal cell microenvironment promotes its apoptosis. Conclusion Compared with leukemic bone marrow stromal cells microenvironment, the bone marrow stromal cell microenvironment of normal bone marrow stromal cells can promote the apoptosis, differentiation and proliferation of Jurkat cells and increase the sensitivity of Jurkat cells to DNR.
