目的探讨大豆异黄酮(ISF)联合顺铂(DDP)化疗对A549肺癌细胞增殖和凋亡的影响。方法采用噻唑蓝(MTT)法观察ISF和DDP联用对A549肺癌细胞增殖的影响,并计算联合指数(Q值)判定联合用药的相互作用,同时采用TUNEL染色法检测二者对A549细胞凋亡的影响。结果 ISF和DDP单独使用均可时间和浓度依赖性地抑制肺癌A549细胞的增殖。DDP浓度在3 mg/L时,ISF和DDP联用后抗细胞增殖作用增强,二者有相加或协同作用,且在一定浓度范围内,联合用药对细胞生长的抑制作用呈量效关系。ISF单用或和DDP联用均可浓度依赖性地诱导肺癌细胞凋亡。结论 ISF可通过诱导A549细胞凋亡而发挥抗肺癌作用;ISF和DDP联用可增强对肺癌A549细胞增殖的抑制和凋亡的诱导,二者具有协同抗肿瘤作用。
Objective To investigate the effects of isoflavone (ISF) and cisplatin (DDP) chemotherapy on the proliferation and apoptosis of A549 lung cancer cells. Methods MTT method was used to observe the effect of combination of ISF and DDP on the proliferation of A549 lung cancer cells. The interaction between the combination of ISF and DDP was evaluated. The apoptosis of A549 cells was detected by TUNEL staining Impact. Results Both ISF and DDP alone could inhibit the proliferation of lung cancer A549 cells in a time and concentration dependent manner. DDP concentration of 3 mg / L, ISF and DDP combined anti-cell proliferation increased, both additive or synergistic effect, and in a certain concentration range, the combination of drugs on cell growth inhibition was dose-effect relationship. ISF alone or in combination with DDP can induce lung cancer cell apoptosis in a concentration-dependent manner. Conclusions ISF can play an anti-lung cancer effect by inducing apoptosis of A549 cells. Combination of ISF and DDP can enhance the inhibition of proliferation and induce the apoptosis of A549 cells. Both of them have synergistic anti-tumor effect.
