目的:建立以微乳为流动相的微乳液相色谱法(MELC),同时测定虎杖中3种主要成分的含量。方法:色谱柱为Hypersil ODS柱(250mm×4mm,5μm),流动相为O/W型微乳1.65%(w/v)SDS-3.3%(v/v)正丁醇-0.3%(v/v)正辛烷-水,检测波长299nm。结果:虎杖苷、白藜芦醇、大黄素分别在(11.3~113.2),(3.4~68.0),(3.8~38.0)mg·L-1范围内与峰面积呈良好线性关系,平均加样回收率和RSD分别为96.0%,1.58%;98.8%,1.37%;101.6%,1.84%。结论:该方法简单,准确,重现性好,可用于虎杖的质量控制。
OBJECTIVE: To establish microemulsion phase chromatography (MELC) with microemulsion as the mobile phase and simultaneously determine the contents of three main components in Polygonum cuspidatum. Method: The column was Hypersil ODS column (250mm×4mm, 5μm). The mobile phase was O/W microemulsion 1.65% (w/v) SDS-3.3% (v/v) n-butanol-0.3% (v/ v) n-octane-water, detection wavelength 299 nm. RESULTS: Polydatin, resveratrol and emodin showed a good linear relationship with the peak area in the range of (11.3–113.2), (3.4–68.0), (3.8–38.0) mg·L-1, respectively. The rate and RSD were 96.0%, 1.58%, 98.8%, 1.37%, 101.6%, and 1.84%, respectively. Conclusion: This method is simple, accurate and reproducible. It can be used for quality control of Polygonum cuspidatum.
