用APAAP免疫组化法检测了高温、化疗药及高温合并化疗药,对人结肠癌细胞THC8908谷胱甘肽转移酶基因表达的影响。发现:正常培养条件下该细胞谷胱甘肽转移酶染色为低度阳性,当一次性给与高温42.5℃1h或5-FU,吗特灵或高温合并5-FU,吗特灵处理后,染色强度不变。但经连续3次上述处理后,单纯给药组呈中度阳性,而且部分高温组细胞均为阴性染色。细胞毒实验亦证明高温可提高化疗药对该细胞的杀伤性。本研究表明,高温可能通过抑制谷胱甘肽转移酶而降低肿瘤细胞的耐药性。
APAAP immunohistochemistry was used to detect the effect of high temperature, chemotherapeutic drugs and high temperature combined with chemotherapy drugs on the expression of glutathione transferase gene of human colon cancer cells THC8908. Found: Under normal culture conditions, the cells were low-positive for glutathione transferase staining. When administered at a high temperature of 42.5°C for 1 hour or 5-FU at a single time, there was a combination of 5-methoxytry (MT) or high temperature with 5-FU. After that, the staining intensity does not change. However, after three consecutive treatments, the single administration group showed moderate positive results, and some high temperature group cells were negatively stained. Cytotoxicity experiments have also demonstrated that elevated temperatures can increase the killing of chemotherapeutic drugs against the cell. This study shows that high temperatures may reduce the resistance of tumor cells by inhibiting glutathione transferase.
