目的探讨稳定高表达X连锁的凋亡抑制蛋白相关因子1(XAF1)基因对肺癌细胞生物学特性的影响。方法使用慢病毒作为载体将XAF1基因转染到肺腺癌A549细胞,用灭瘟素试剂(blasticidin)进行筛选,建立稳定高表达XAF1基因细胞株A549,用Western blot法检测细胞生存素蛋白和上皮细胞钙黏蛋白表达的变化。结果成功构建稳定高表达XAF1基因肺癌细胞株A549。A549中XAF1及上皮细胞钙黏蛋白表达水平升高(P<0.05),细胞生存素蛋白表达下降(P<0.05)。结论用慢病毒载体可成功构建稳定表达XAF1基因的肺腺癌细胞株A549。XAF1基因高表达可抑制肺癌细胞的生长和转移。
Objective To investigate the effect of stable and highly expressed X-linked inhibitor of apoptosis-related protein 1 (XAF1) gene on the biological characteristics of lung cancer cells. Methods XAF1 gene was transfected into lung adenocarcinoma A549 cells using lentivirus as a vector and screened with blasticidin to establish a stable and highly expressed XAF1 cell line A549. The expression of survivin and epithelial cells Changes in cell cadherin expression. Results The stable and highly expressed XAF1 lung cancer cell line A549 was successfully constructed. The A549 expression of XAF1 and E-cadherin increased (P <0.05), while the expression of survivin protein decreased (P <0.05). Conclusion The lung adenocarcinoma cell line A549 stably expressing XAF1 gene can be successfully constructed by lentiviral vector. High expression of XAF1 gene can inhibit the growth and metastasis of lung cancer cells.
