目的:探讨化瘀理肺方对博莱霉素致大鼠肺间质纤维化的治疗作用及机制。方法:将大鼠随机分为对照组、模型组、中药组、激素组,采用博莱霉素经气管内插管注入方法(5mg/kg)建立肺间质纤维化模型,对照组注入生理盐水(0.25mL/只)作为正常对照,从造模后的第2天起中药组给予化瘀理肺方药物灌胃(13410mg/kg);对照组、模型组予生理盐水灌胃(2mL/只);激素组予氢化可的松琥珀酸钠腹腔注射(25mg/kg),于给药后的第14天、28天分别处死,取肺组织行苏木素-伊红(HE)染色观察病理变化情况,免疫组化法测定肺组织中Smad3、Smad7蛋白的表达情况以及应用凝胶酶谱法检测MMP-2/9的活性。结果:中药组可见炎性细胞较少,肺泡结构基本正常,肺泡炎与肺纤维化较轻。中药组Smad3蛋白表达明显低于模型组和激素组;Smad7蛋白表达明显高于模型组和激素组;中药组MMP-2/9的活性明显低于模型组和激素组。结论:化瘀理肺方可明显减轻大鼠肺泡炎和肺纤维化的程度,可能通过Smad信号通路从而影响MMP-2/9的活性发挥作用。
Objective: To investigate the therapeutic effect of Huayu Lifang Fang on bleomycin-induced pulmonary interstitial fibrosis in rats and its mechanism. Methods: The rats were randomly divided into control group, model group, traditional Chinese medicine group and hormone group. Bleomycin was used to establish pulmonary interstitial fibrosis model by intratracheal intratracheal instillation (5mg / kg). The control group was injected with normal saline (0.25mL / only) as normal control, from the second day after modeling, the traditional Chinese medicine group was administered with Huayu Lifei Recipe (13410mg / kg); in the control group, the model group was given saline ). The corticosteroid group was intraperitoneally injected with hydrocortisone succinate (25mg / kg). The rats in the hormones group were sacrificed on the 14th day and the 28th day after the administration. The lung tissues were stained with hematoxylin and eosin (HE) to observe the pathological changes The expression of Smad3 and Smad7 protein in lung tissue was detected by immunohistochemistry and the activity of MMP-2/9 was detected by gel enzyme-linked immunosorbent assay. Results: There were fewer inflammatory cells, alveolar structure and alveolitis in the Chinese herbal medicine group. The expression of Smad3 protein in traditional Chinese medicine group was significantly lower than that in model group and hormone group; the expression of Smad7 protein was significantly higher than that in model group and hormone group; the activity of MMP-2/9 in Chinese herbal medicine group was significantly lower than that in model group and hormone group. Conclusion: HuaYuLiFang can obviously reduce the degree of alveolitis and pulmonary fibrosis in rats, which may affect the activity of MMP-2/9 through the Smad signaling pathway.
