目的探讨二烯丙基二硫(DADS)诱导人鼻咽癌CNE1细胞凋亡的生物学效应。方法通过MTT法检测DADS对CNE1细胞生长的影响;用光学显微镜、流式细胞仪研究DADS诱导细胞凋亡的作用。免疫组织化学法检测细胞内Bcl-2、Bax蛋白的表达,Western blot法检测细胞内Caspase-3蛋白的表达。结果DADS能抑制CNE1细胞的生长。DADS处理CNE1细胞后,细胞呈凋亡特征性改变。流式细胞分析显示DADS呈浓度依赖性诱导CNE1细胞凋亡。免疫组织化学结果表明DADS处理细胞后,Bax蛋白表达升高,Bcl-2蛋白表达下降。随药物浓度的增加Caspase-3蛋白表达增强。结论DADS具有诱导CNE1细胞凋亡的作用,其机制可能与Bcl-2/Bax下降,促进Caspase-3活化有关。
Objective To investigate the biological effects of diallyl disulfide (DADS) on apoptosis of human nasopharyngeal carcinoma CNE1 cells. Methods The effect of DADS on the growth of CNE1 cells was detected by MTT assay. The effect of DADS on apoptosis was investigated by light microscopy and flow cytometry. Immunohistochemistry was used to detect the expression of Bcl-2 and Bax in cells, and Western blot was used to detect the expression of Caspase-3 protein. Results DADS can inhibit the growth of CNE1 cells. After DADS treatment of CNE1 cells, the cells showed characteristic changes of apoptosis. Flow cytometry analysis showed that DADS induced CNE1 cell apoptosis in a concentration-dependent manner. Immunohistochemistry showed that after DADS treatment, the expression of Bax protein and Bcl-2 protein decreased. Caspase-3 protein expression increased with increasing drug concentration. Conclusion DADS can induce the apoptosis of CNE1 cells. The mechanism may be related to the decrease of Bcl-2 / Bax and the activation of Caspase-3.
