[目的]应用流式荧光杂交法检测人乳头瘤病毒(HPV),分析宫颈疾病患者HPV感染及分型情况。[方法]以2013年1月至2013年12月浙江省宁海县第一医院4 041例宫颈疾病患者作为研究对象,采集宫颈脱落细胞,用流式荧光杂交技术检测HPV基因型,并对HPV感染率、基因亚型、感染类型和年龄分布等进行分析。[结果]检出HPV感染744例(18.41%),其中单一亚型感染603例(14.92%),多重亚型感染141例(3.49%)。可供检测的26种HPV基因亚型均有检出,最常见的亚型为HPV-16、52、58、39、18、61、59、6等,其中低危型以HPV-61、6、11亚型感染为主。高危型以HPV-16、52、58亚型感染为主。HPV感染亚型以高危亚型为主,单一高危亚型感染阳性率13.07%(528/4 041),多重高危亚型感染阳性率2.15%(87/4 041)。31~40岁妇女HPV感染阳性率最高,阳性率22.22%,50岁以上者HPV感染阳性率最低,阳性率13.99%,且各年龄组中均以高危亚型感染检出率最高。[结论]宫颈疾病患者中HPV感染率较高,基因型分布较广,流式荧光杂交法是检测HPV感染的快速有效方法,HPV分型检测对宫颈疾病的防治意义重大。
[Objective] To detect human papillomavirus (HPV) by flow cytometry (FCM) and analyze HPV infection and typing in patients with cervical diseases. [Methods] From April 2013 to December 2013, 4 041 cervical disease patients in the First Hospital of Ninghai County of Zhejiang Province were used as research objects to collect the exfoliated cervical cells. The HPV genotypes were detected by flow cytometry and HPV infection Rate, genotype, infection type and age distribution. [Results] 744 cases (18.41%) were detected with HPV infection, of which 603 cases (14.92%) were single subtypes and 141 cases (3.49%) were multiple subtypes. The 26 most common HPV subtypes tested were HPV-16, 52, 58, 39, 18, 61, 59, 6, among which HPV-61,6 , 11 subtype infection. High-risk type to HPV-16, 52,58 subtype infection-based. The HPV subtypes were mainly high risk subtypes. The positive rate of single high risk subtypes was 13.07% (528/4 041) and multiple high risk subtypes was 2.15% (87/4 041). The positive rate of HPV infection was the highest among women aged 31-40, with a positive rate of 22.22%. The positive rate of HPV infection was the lowest in those over 50 years old, with a positive rate of 13.99%. The detection rate of high-risk subtypes was the highest among all age groups. [Conclusion] The prevalence of HPV in cervical disease patients is high and the genotypes are widely distributed. Flow-fluorescent hybridization is a rapid and effective method to detect HPV infection. The detection of HPV typing is of great significance for the prevention and treatment of cervical diseases.
