目的 :研究SMU1(自制抗CD3)对正常人外周血单个核细胞 (PBMNC)的体外功能效应。方法 :①以SMU16种不同浓度、IL 2、MCD3(丝裂性抗CD3)孵育PBMNC ,观察细胞密度和形态变化 ,MTT法测定细胞增殖力 ;②分别用SMU10 .5mg/L、5mg/L、IL 2 2 0 0kU/L培养PBMNC 3d ,ELISA法测定上清液IL 6、IL 8、IL 12、IFN α、TNF α及G CSF水平。结果 :IL 2组和MCD3组细胞增多成团 ,细胞异形性明显 ,增殖力显著高于实验对照组。而SMU1各组细胞形态、密度无明显变化。PBMNC经SMU1孵育后上清IL 8、IL 12、IFN α水平显著升高 ,经IL 2孵育后上清IL 8、TNF α、IFN α水平升高。结论 :SMU1无丝裂作用 ,但对PBM NC分泌细胞因子有重要影响 ,其促分泌作用与IL 2有所不同
AIM: To investigate the in vitro functional effects of SMU1 (self-made anti-CD3) on peripheral blood mononuclear cells (PBMNC) from healthy donors. Methods: (1) PBMNCs were cultured in different concentrations of SMU at different concentrations of IL2 and MCD3 (mitogenic anti-CD3) to observe the cell density and morphological changes. Cell proliferation was measured by MTT assay. The PBMNCs were cultured with IL 2 0 0 kU / L for 3 days, and the levels of IL 6, IL 8, IL 12, IFN α, TNF α and G CSF were measured by ELISA. Results: The cells in IL 2 group and MCD3 group increased into lumps, and the cell atypia was obvious. The proliferation ability was significantly higher than that of the experimental control group. The SMU1 cells in each group morphology, density no significant change. The levels of IL 8, IL 12 and IFN α in supernatant of PBMNC after SMU1 incubation were significantly increased, and the levels of IL 8, TNF α and IFN α in supernatant of PBMNC increased after incubation with IL 2. CONCLUSIONS: SMU1 has no mitosis, but has an important effect on the secretion of cytokines by PBM NC, and its secretion is different from that of IL2
