人 B1结合因子(human B1 binding factor, hB1F)是本实验室克隆到的一个新的转录因子,它能够结合于HBV增强子Ⅱ(EN Ⅱ)的B1区并反式激活其功能.对hB1F和其他反式激活EN Ⅱ的肝细胞核因子间在功能上的相互关系进行了研究.氯霉素乙酰转移酶(chloramphenicolacetyltransferase, CAT)报告基因分析发现, hB1F与 HNF3β, HNF4间在激活 EN Ⅱ的功能上仅存在简单的加和性,而hB1F和HNF1共同存在的情况下却能够产生很强的协同效应.进一步利用GSTpull-down实验在体外证明了这两种因子间存在特异的直接相互作用.研究结果表明,hB1F与HNF1协同地调控HBV增强子Ⅱ的功能.
Human B1 binding factor (hB1F) is a new transcription factor cloned in our laboratory. It can bind to the B1 region of HBV enhancer Ⅱ (EN Ⅱ) and transactivate its function. The functional relationship between hB1F and other trans-activated EN II hepatocytes was studied. Chloramphenicolacetyltransferase (CAT) reporter gene analysis revealed that there was only simple additive function between hB1F and HNF3β and HNF4 in the activation of EN Ⅱ, but the combination of hB1F and HNF1 could produce very good Strong synergy. Further use of GSTpull-down experiments in vitro proved that there is a specific direct interaction between these two factors. The results show that hB1F and HNF1 synergistically regulate the function of HBV enhancer Ⅱ.
