目的 探讨香烟提取物(CSE)对人支气管上皮细胞表达人β防御素2(hBD2)的影响。方法利用不同浓度(4%和8%)的CSE刺激人支气管上皮细胞(16HBE),分别于0、6、12、24、48 h收集细胞总蛋白,运用酶联免疫吸附法(ELISA)测定hBD2的蛋白表达水平。结果 4%和8%CSE均诱导16HBE于6、12、24、48 h进行性hBD2表达下降。8%CSE干预组变化幅度大于4%CSE干预组。结论 CSE抑制人支气管上皮细胞表达hBD2,此机制可能参与香烟对气道防御的抑制作用。
Objective To investigate the effect of cigarette smoke extract (CSE) on human β-defensin 2 (hBD2) expression in human bronchial epithelial cells. Methods Human bronchial epithelial cells (16HBE) were stimulated with CSE at different concentrations (4% and 8%). Total cellular proteins were collected at 0, 6, 12, 24 and 48 hours, respectively, and hBD2 was detected by enzyme-linked immunosorbent assay Protein expression level. Results Both 4% and 8% CSE induced 16HBE decreased hBD2 expression at 6, 12, 24 and 48 h. 8% CSE intervention group changed more than 4% CSE intervention group. Conclusion CSE inhibits the expression of hBD2 in human bronchial epithelial cells. This mechanism may be involved in the inhibitory effect of cigarette on airway defense.
