目的采用MTS比色法检测非小细胞肺癌(NSCLC)患者癌性胸水中肿瘤细胞对常用化疗药物的敏感性。方法采用密度梯度离心法提取19例NSCLC患者癌性胸水中的肿瘤细胞,应用MTS比色法检测其对顺铂(DDP)、多西他赛(DOC)、长春瑞滨(NVB)、氟尿嘧啶(5-FU)、博来霉素(BLM)、奈达铂(NDP)、吉西他滨(GEM)、培美曲塞(MTA)等8种化疗药物的敏感性,以及这8种化疗药物对NSCLC胸水细胞的抑制作用。结果 19例NSCLC患者胸水标本中肿瘤细胞均成功提取。各化疗药物对NSCLC抑制作用之间差异均有统计学意义(P<0.001),从高到低依次为:NDP>DDP>GEM>NVB>5-FU>MTA>DOC>BLM。随着药物浓度的增加,各化疗药物对肿瘤细胞的抑制作用逐渐增大,癌性胸水中肿瘤细胞对各化疗药物的敏感性增加。结论 MTS比色法作为一种简便、快速、安全、灵敏性高的体外药敏试验方法,可用于NSCLC患者的个体化治疗选择合适的化疗药物。
Objective To detect the sensitivity of chemotherapeutic drugs to tumor cells in cancerous pleural effusion by MTS colorimetric assay in patients with non-small cell lung cancer (NSCLC). Methods Tumor cells from cancerous pleural effusion of 19 patients with NSCLC were collected by density gradient centrifugation. The expression of DDP, DOC, NVB and Fluorouracil 5-FU, BLM, NDP, GEM and MTA, as well as the sensitivity of these 8 chemotherapeutic drugs to NSCLC pleural effusion Cell inhibition. Results Tumor cells from 19 cases of NSCLC were successfully extracted. The chemotherapeutic agents showed significant differences (P <0.001) in the inhibition of NSCLC. The descending order was NDP> DDP> GEM> NVB> 5-FU> MTA> DOC> BLM. With the increase of drug concentration, the inhibitory effect of each chemotherapeutic drug on tumor cells gradually increased, and the sensitivity of tumor cells in cancerous pleural effusion to each chemotherapeutic drug increased. Conclusion As a simple, rapid, safe and sensitive in vitro susceptibility testing method, MTS colorimetric assay can be used to select suitable chemotherapeutic drugs for individualized treatment of NSCLC patients.
