目的 :观察甾体皂苷化合物 (化合物 9714 )对血栓形成、血小板聚集及血液流变学的影响。方法 :采用动静脉旁路血栓形成模型 ,测定化合物 9714对血栓形成的影响 ;采用比浊法 ,测定其对大鼠血小板聚集的影响 ;采用大鼠急性血淤模型 ,测定其对血液流变学的影响。结果 :化合物 9714 10 ,2 0 ,4 0mg/kg组均能明显减轻血栓的干重及湿重 ;可抑制ADP、胶原诱导的大鼠血小板聚集 ;降低急性血淤大鼠的全血黏度、血浆黏度、红细胞聚集性 ,以及增强红细胞变形性。结论 :化合物 9714通过抑制血小板聚集、改善血液流变学来发挥其抗血栓形成作用。
AIM: To investigate the effect of steroidal saponin compound (compound 9714) on thrombus formation, platelet aggregation and hemorheology. METHODS: The model of arteriovenous bypass thrombosis was established to determine the effect of compound 9714 on thrombosis. The turbidimetric method was used to determine the effect of compound 9714 on platelet aggregation in rats. Acute blood stasis model was used to determine the effect of compound 9714 on blood rheology Impact. Results: Compound 9714 10, 20, 40 mg / kg significantly reduced thrombus dry weight and wet weight, inhibited ADP and collagen-induced platelet aggregation in rats, decreased whole blood viscosity, Viscosity, erythrocyte aggregation, and enhanced erythrocyte deformability. Conclusion: Compound 9714 exerts its antithrombotic effect by inhibiting platelet aggregation and improving hemorrheology.
