目的应用miRNA芯片技术研究小分子RNA(miRNA)对犬心房纤颤(AF)的调节作用,发现了AF的新机制,为新药研究提供了新的靶点。方法将杂种犬随机分为两组,假手术组(n=4);利用高频起搏器对右心耳进行刺激起搏诱发持续性AF组(n=5),取左心房组织常规抽提总RNA,采用含有468种miRNA的芯片检测系统进行miRNA表达谱差异分析。结果和假手术组相比,AF模型组的miRNA表达谱中差异表达的miRNA共有10个,其中有4个miRNA上调,6个miRNA下调。结论miRNA参与了持续性AF的调节作用,提示miRNA是抗心律失常药物作用的新靶点。
Objective To study the regulatory effect of small RNA (miRNA) on atrial fibrillation (AF) in dogs by using miRNA microarray technology and to find out a new mechanism of AF, which provides a new target for new drug research. Methods The mongrel dogs were randomly divided into two groups: sham operation group (n = 4); continuous AF group (n = 5) Total RNA was analyzed using a microarray detection system containing 468 miRNAs for miRNA expression profiling. Results There were 10 miRNAs differentially expressed in the AF model group compared with the sham-operation group, of which 4 miRNAs were up-regulated and 6 miRNAs were down-regulated. Conclusion miRNA participates in the regulation of persistent AF, suggesting that miRNAs are new targets of antiarrhythmic drugs.
